Abstract
Research has been carried out on the isolation and characterization of secondary metabolites compounds of plant stem bark Niko (Grewia koordersiana Burret) from Camplong, Kupang district. The purpose of this research is to know the types of secondary metabolites and characteristics of secondary metabolites compounds of stem bark of Niko plants. The method used is extraction, chromatography and spectrophotometry. A total of 500 g of Niko bark powder, macerated with methanol solvent for 24 hours and obtained a thick methanol extract of Niko bark weighing 260.45 g with a yield of 52.09%. The phytochemical test results showed positive flavonoid, alkaloid and saponin compounds. Fractionation of the active compound separation using column chromatography with the mobile phase of methanol: ethyl acetate (1: 3 v / v) and obtained 20 fractions. The TLC test results on all fractions with eluent methanol: ethyl acetate (1: 3 v / v) so as to produce 4 fractions which have a single spot namely fraction 11, 16 (0.3) and fraction 12, 15 (0.7). The two methanol extract isolates of Niko bark were identified by UV-Vis spectrophotometry and showed two maximum absorption peaks, namely at wavelengths of 280 nm (band I) and 235 nm for fraction A (11.16), while for fraction B (12,15) maximum absorption occurs at wavelength 280 nm (band I) and 245 nm (band II). The transition of electrons at wavelengths 235 and 245 nm (band I) indicates a transition *. Whereas at 280 nm wavelength (Ribbon II) shows a transition n *. The results of the FTIR spectrum in fractions A and B showed the presence of stretching OH phenol vibrations (3383.14 cm-1 ), aliphatic C-H (2983.88 cm-1 ), C=O isolated carbonyl (1741.72 cm-1 ), C=C aromatic (1375.25 cm-1 ) and alcohol C-O (1244.09 cm-1 , 1045.42 cm-1 , 1033.85 cm-1 ). Based on the results of the UV-Vis and IR spectrum, the two isolates contained flavonoids of flavanone and flavanonol types.
