Abstract

Research has been carried out on the isolation and characterization of alkaloid compounds from the methanol extract of the leaves of the amethyst plant (Datura metel L) from Pakubaun village, east Amarasi, Kupang district. This study aims to determine the characteristics of alkaloid compounds from the leaves of the amethyst plant. The method used is extraction, chromatography, and spectrophotometry. A total of 200 g amethyst plant leaf powder, was accelerated with methanol solvent for 24 hours. The results obtained from the thick methanol extract of amethyst plant leaves weighing 28.9 g with a yield of 14.09%. Phytochemical test results showed positive containing alkaloids, flavonoids, steroids, and saponins. The separation of active compounds by fractionation, using column chromatography with the mobile phase of petroleum ether: chloroform (9: 1; 8: 2; 7: 3 v / v) and obtained 39 fractions. TLC test results on all fractions and those that have a single spot are fractions 19, 20, 21, 22 (0.42) and fractions 23, 24, 25 (0.32). Both isolates of fraction A and fraction B of the amethyst plant leaves were identified by UV-Vis spectrophotometry producing a maximum wavelength of 260 nm in fraction A while for fraction B yielding a maximum wavelength of 275 nm. Analysis using FTIR in isolates of fraction A and fraction B indicates the presence of alkaloid compounds that have the characteristics of aliphatic CH, CH3, C = O, and C-N functional groups.
