Abstract
The final disposal site (TPA) in Kupang City is located in Alak District with an area of 9.14 Ha and has been operating since 1998. The operation of Alak TPA uses the controlled landfill method which is an improvement from open dumping with the aim of reducing the potential for environmental disturbances caused. In its implementation, waste that enters the TPA does not go through a sorting and recycling process, but is directly dumped into a waste disposal area which is then covered with white soil and pressed with heavy equipment. The method used in this study is the pour plate method or sprinkling method. The pour plate method or sprinkling method is a method for obtaining pure cultures from a mixed population of microorganisms by diluting the specimen which is then poured into a sterile cup followed by pouring the agar medium that has been melted and cooled (at a temperature of ±500°C). The results of the study obtained at the Final Disposal Site in Alak District, Kupang City successfully identified 3 types of aerobic bacteria, namely isolate BTB1 has similarities with the genus Azothobacter, BTB2 has similarities with the genus Azomonas, and BTB3 has similarities with the genus Xanthobacter. Isolate bacteria BTB1 has similarities with the genus Azothobacter, coccus cell shape, gram negative, yellow color, convex elevation, wavy colony edges and coccus shape. Requires O2, catalase positive, ferments glucose, lactose, fructose sugars, grows at temperatures of 15°, 30°, and 50°, can grow at pH ranges of 4 and 9, grows at salt concentrations of 5% and 15%. Isolate bacteria BTB2 which has similarities with the genus Azomonas, coccus shape, gram negative. yellow colony color, pulvinate elevation, entire colony edge, coccus colony shape, requires positive O2, catalase positive, ferments glucose, lactose, fructose sugars, grows at temperatures of 15°, 30°, and 50°. grows at pH ranges of 4 and 9, grows at 5% and 15% salt. Isolate of BTB3 bacteria that has similarities with the genus Xanthobacter coccus shape, gram negative, white colony color, convex elevation, entire colony edge, coccus colony shape, requires O2, catalase positive, ferments glucose, lactose, fructose sugars, grows at temperatures of 15°, 30°, and 50, grows at pH ranges of 4 and 9, grows at salt concentrations of 5%, 10%, and 15%.
