Abstract

This study aims to determine the characteristics of alkaloid compounds from the leaves of the amethyst plant. The method used is extraction, chromatography, and spectrophotometry. A total of 200 g amethyst plant leaf powder, was accelerated with methanol solvent for 24 hours. The results obtained from the thick methanol extract of amethyst plant leaves weighing 28.9 g with a yield of 14.09%. Phytochemical test results showed positive containing alkaloids, flavonoids, steroids, and saponins. The separation of active compounds by fractionation, using column chromatography with the mobile phase of petroleum ether: chloroform (9: 1; 8: 2; 7: 3 v / v) and obtained 39 fractions. TLC test results on all fractions and those that have a single spot are fractions 19, 20, 21, 22 (0.42) and fractions 23, 24, 25 (0.32). Both isolates of fraction A and fraction B of the amethyst plant leaves were identified by UV-Vis spectrophotometry producing a maximum wavelength of 260 nm in fraction A while for fraction B yielding a maximum wavelength of 275 nm. Analysis using FTIR in isolates of fraction A and fraction B indicates the presence of alkaloid compounds that have the characteristics of aliphatic CH, CH3, C = O, and C-N functional groups.
