Abstract

Springs are water sources that appear on the surface of the ground. The existence of springs is crucial for the community because it is used for basic human needs. This study aimed at determining the types and the characteristics of the bacteria in the Oe Kiu spring, Soba Village, West Amarasi District, Kupang Regency. The working procedures included preparation, testing, and data analysis. The preparation consisted of sterilizing the instrument, making NA media, and taking water samples. The test consisted of bacterial culture, bacterial purification, and bacteria identification. The identification stage included macroscopic,microscopic, and biochemical observations. Furthermore, the data obtained were analyzed descriptively and qualitatively. Then the data were displayed in tables and figures. The results showed that the characteristics of bacteria that were mostly found in Oe Kiu springs were bacilli and coccus. These bacteria had different characteristics. Of the six bacterial genera isolates that were identified, where these five isolates have closenees or similarity to the bacterial genera were identified. They were Bacillus, Acetobacter, Alcaligenes, Vibrio, and Escherichia.
