Abstract
Salome is one of the processed meat-based food products that become a favorite snack for all of people in Kupang City. As a food, salome could be contaminated by Escherichia coli bacteria. Escherichia coli are one of the bacteria that are susceptible to contaminating salome and could cause diarrhea or foodborne diseases due to poor food hygiene. The process of bacterial contamination can occur due to several factors. This study aims to determine the presence of Escherichia coli bacteria in processed meat products in Kupang City. This research is a descriptive research with purposive sampling technique. Isolation of bacteria using EMBA media, population calculations were carried out by the cup counting method, namely a dish with a number of colonies from 25 to 250 was selected and then the number of each colony in the dish was calculated based on its morphological appearance. Macroscopic characterization was carried out by observing the morphology of the different colonies on the plate to obtain Escherichia coli bacteria isolates. The microscopic characteristics were carried out using the Gram stain method and observed using a light microscope. The results showed that the population of Escherichia coli bacteria from a 10-3 dilution in Taman Nostalgia was 8.4 x 104 CFU / ml, SMPN 1 Kupang was 8.4 x 104 CFU / ml, SDK St. Yoseph 1 Kupang ie 5 x 104 CFU / ml and SMAN 1 Kupang 1.11 x 105 CFU / ml. The macroscopic characterization at the four locations had the same shape, elevation, and edge. Microscopic characteristics were carried out using the gram staining method with the results that all bacterial isolates were gram-negative bacteria that had a coccus cell shape and single cell arrangement. Based on the results of the study, it can be concluded that the population of Escherichia coli bacteria can be found in salome from Taman Nostalgia, SMPN 1 Kupang, SDK St. Yoseph 1 Kupang, and SMAN 1 Kupang. The isolated Escherichia coli bacteria have the same characteristics both macroscopically which include the same shape, elevation, and edges, while microscopically it shows that all Escherichia coli bacteria have gram negative, coccus cell shape, and single cell arrangement.
