Abstract
Waste is the residual disposal that results from a production process both domestic (household) and industry that can pollute the environment, one of them is dye waste which is produced from ikat woven crafts such as those in Ina Sabu ikat. Dyes in textiles are one of the non-biodegradable organic pollutants which means they cannot be destroyed or broken down by living organisms. Therefore, treatment of waste produced to reduce the potential for environmental pollution is needed, namely biological treatment through biodegradation processes using Lignolytic bacteria. This bacterium is able to degrade dye waste because it has an extracellular enzyme that is able to degrade dye waste. This research is for the purpose to see the potential of Lignolytic bacteria in the degradation of the dye waste. Bacterial isolation was carried out by enrichment culture (enrichment) techniques on MSM (Mineral Salt Medium) media. From the isolation results, 17 bacterial isolates were then selected for Lignolytic activity by point inoculation method on NA medium containing 1% tannic acid. The results obtained were 8 isolates with lignolytic ability which were marked by the formation of a brown zone around the colony. Then 3 isolates were taken which have the biggest brown zone to be observed macroscopically, microscopically, testing biochemical properties, and testing physiological properties. Determination of the bacterial genus is done by comparing the results of characterization with Bergeys's Manual of Determinative Bacteria 9th there are three genera that have similarities to the genera Acetobacter, Micrococcus and Azotobacter. Of the 3 bacterial isolates, their ability to degrade sulfur dyes was tested by inoculating bacterial isolates into NB 0.4% containing dye waste. The ability of bacteria to degrade the dye waste can be seen from the change in color in the media and was measured by a spectrophotometer with a wavelength of 354 nm which was carried out for 48 hours every 24 hours. These three isolates are able to degrade sulfur coloring waste.
